Resumen El objetivo de esta investigación, fue determinar la incidencia y distribución de la enfermedad mano de chango en maíz en nueve estados de la República Mexicana (Tlaxcala, Puebla, Edo. de México, Hidalgo, Veracruz, Morelos, Oaxaca, Chiapas y Jalisco). Los sitios de evaluación se georreferenciaron, se estimó la incidencia y se realizó una encuesta con los productores para determinar la identidad del germoplasma utilizado. La mano de chango se encontró afectando plantas de maíz en localidades de los estados de Puebla con incidencia de 24%, Hidalgo (17.05%), Morelos (11.7%), Chiapas (7.28%), Tlaxcala (7.0%), Oaxaca (6.5%), Veracruz (6.3%), Edo. de México (6.2%), y Jalisco (3.0%). Los datos mostraron una correlación baja, pero positiva (r=0.30) entre la altitud y la incidencia. Observaciones indicaron que el germoplasma nativo es más susceptible a la enfermedad que el mejorado.

Abstract The incidence of maize monkey´s hand disease was determined in nine states of the Mexican Republic. The evaluation sites were georeferenced, and incidence was estimated along with the farmers to determinate the identity of the germplasm used. Monkey´s hand was found affecting maize plants in localities of the States of Puebla with an incidence of 24%, Hidalgo (17.05%), Morelos (11.7%), Chiapas (7.28%), Tlaxcala (7.0%), Oaxaca (6.5%), Veracruz (6.3%), Mexico (6.2%), and Jalisco (3.0%). Data showed a low but positive correlation (r= 0.30) between altitude and incidence. Native germplasm is more susceptible to the monkey´s hand disease than improved germplasm.

RESUMO O virus do tomate rugoso marrom (tomato brown rugose fruit virus - ToBRFV) é um vírus emergente que ameaça a produção de tomate e pimenta em diferentes países. Este trabalho teve como objetivo avaliar o efeito da infecção do ToBRFV na morfologia vegetal e na qualidade do fruto em oito variedades de pimenta malagueta e propor uma escala diagramatica para avaliar a severidade em folhas infectadas considerando a porcentagem de área foliar afetada. Dezesseis tratamentos foram testados, que incluíram oito variedades de pimentão e duas condições experimentais (inoculadas e não inoculadas). Cinco dias após o transplante, plantas de cada variedade de pimenta foram inoculadas mecanicamente com ToBRFV e as variáveis morfológicas e de qualidade dos frutos foram avaliadas. A comparação das médias de cada variedade inoculada versus não inoculada foi realizada com o teste t de Student (P ≤ 0,05) para grupos independentes. O ToBRFV infectou todas as variedades inoculadas causando lesões necróticas locais, clorose, mosaico, mosqueamento, necrose do caule, ampulação, deformação, enrolamento e necrose da folha; da mesma forma, foram observadas plantas assintomáticas. O ToBRFV afetou significativamente a altura, número de entrenós, número de flores, peso fresco da raiz, peso médio do fruto, número de sementes e diâmetro polar-equatorial do fruto. A variedade de pimenta habanero apresentou a menor severidade da doença, enquanto a maior severidade foi observada na variedade de pimenta serrano ‘Tampiqueño’.

ABSTRACT Tomato brown rugose fruit virus (ToBRFV) is an emerging virus that threatens tomato and chili production worldwide. The objective of this work was to evaluate the effect of ToBRFV infection on plant morphology and fruit quality in eight varieties of pepper and to propose a diagrammatic scale to evaluate severity in infected leaves considering the percentage of affected leaf area. Sixteen treatments were tested, consisting in eight varieties of chili and two experimental conditions (inoculated and non-inoculated). Five days after transplanting, each chili variety was mechanically inoculated with ToBRFV and morphological and fruit quality variables were evaluated. The comparison of means of each inoculated versus non-inoculated variety was performed with the t-Student test (P ≤ 0.05) for independent groups. ToBRFV infected all inoculated plants causing local necrotic lesions, chlorosis, mosaic, mottling, stem necrosis, ampulation, deformation, rolling and leaf necrosis depending of the variety; likewise, asymptomatic plants were observed. ToBRFV significantly affected height, number of internodes, number of flowers, fresh root weight, average fruit weight, number of seeds, and polar-equatorial diameter of the fruit. The habanero chili showed the least severity, while the highest severity was shown in the serrano chili ‘Tampiqueño’.

RESUMEN El Tomato brown rugose fruit virus (ToBRFV) es un virus emergente que amenaza la producción de tomate y chile en todo el mundo. Esta investigación tuvo como objetivo evaluar el efecto de la infección del ToBRFV en la morfología de la planta y calidad de frutos en ocho variedades de chile y proponer una escala diagramática para evaluar la severidad en hojas infectadas considerando el porcentaje de área foliar afectada. Se probaron 16 tratamientos, que incluyeron ocho variedades de chile y dos condiciones experimentales (inoculado y no inoculado). Cinco días después del trasplante, cada variedad de chile se inoculó mecánicamente con ToBRFV y se evaluaron variables morfológicas y de calidad de frutos. La comparación de medias de cada variedad inoculada versus no inoculada se realizó con la prueba t-Student (P ≤ 0.05) para grupos independientes. El ToBRFV infectó a todas las variedades inoculadas ocasionando lesiones locales necróticas, clorosis, mosaico, moteado, necrosis de tallo, ampulación, deformación, enrollamiento y necrosis de hoja; asimismo, se observaron plantas asintomáticas. El ToBRFV afectó significativamente la altura, número de entrenudos, número de flores, peso fresco de raíz, peso promedio de fruto, número de semillas y diámetro polar-ecuatorial del fruto. La variedad chile habanero mostró la menor severidad, mientras que la mayor severidad se observó en la variedad de chile serrano ‘Tampiqueño’.

Abstract. Recently, an increase in the presence of symptom of a possible new disease on maize has been observed which farmers have called “mano de chango” (monkey´s hand) due to the prolificacy shown by the ears. Initial reports, in the 1960s, indicated, with no proofs, that this disease was caused by fungi, insects, or nutritional deficiencies with incidences up to 100% and losses in grain yield. This is the first work where evaluation of incidence of the disease, in 21 commercial hybrids in the Toluca Valley, State of Mexico, a comercial hybrid and a native variety in the Mezquital Valley, Hidalgo State, and six improved open pollinated in Montecillo, State of Mexico. The highest incidences and grain yield losses were recorded in the genotype Aspros HC8 and the native variety “Hidalgo”, in Mezquital Valley, Hidalgo State, where disease symptoms showed considerable yield reduction. It is necessary to continue studies to determine the etiology and generate information to select genetic resistance to this possible new disease.

Resumen. Recientemente, se ha observado un incremento en la presencia de una posible nueva enfermedad en plantas de maíz que genera prolificidad de mazorcas que los agricultores denominan “mano de chango”. Estos síntomas se han observado en áreas tropicales, subtropicales y del altiplano de México. Observaciones iniciales, en los años 1960s, indicaron, sin pruebas, que esta enfermedad era causada por hongos, insectos, o deficiencias nutricionales con incidencia de hasta 100% y pérdidas en producción de grano. Este es el primer trabajo en que se evalúa la incidencia de la enfermedad en 21 híbridos comerciales en el Valle de Toluca, México, un híbrido comercial y una variedad nativa de polinización libre en el Valle del Mezquital, Hidalgo, y seis variedades mejoradas de polinización libre en Montecillo, Texcoco, México. La mayor incidencia y pérdidas en rendimiento de grano por la enfermedad se observó en los genotipos Aspros HC8 y la variedad nativa “Hidalgo”, en el Valle del Mezquital, Hidalgo, en donde la presencia de síntomas de la enfermedad mostró una considerable reducción en rendimiento. Es necesario continuar estudios para investigar su etiología y generar información para selección de resistencia genética a esta posible nueva enfermedad.

Abstract Mycobacterium avium subsp. paratuberculosis (MAP) is the causative agent of paratuberculosis (PTB), disease that causes a syndrome of bad nutrient absorption, weight loss and eventually death. The intestine is the main target organ where the infection develops; however, there is evidence of infection by MAP in extra-intestine sites of sheep, including mesenteric nodes and semen. The aim of the study was to identify the presence of MAP in reproductive tissue and semen of infected Pelibuey rams in clinical state of PTB. Seven rams were used in clinical PTB state and a non-infected ram by MAP of the Pelibuey breed, confirmed by serology, nPCR and bacteriological culture, with average weight and age of 57.23 ± 1.73 kg and 2.91 ± 0.17 years, respectively. The presence of MAP was identified in different tissue samples: spleen (1/7, 14.3% and 2/7, 28.6%), small intestine (3/7, 42.9% and 4/7, 57.1%) and mesenteric lymph nodes (3/7, 42.9% and 3/7, 42.9%), with nPCR and culture, respectively. It was also identified in epididymis tissue (1/7, 14.3%), Cowper gland (2/7, 28.6%) and prostate (1/7, 14.3%), using nPCR, although without detection in culture. It was identified in testicular tissue in 42.8% (3/7; culture or nPCR technique), but in 28.6% (2/7) with both techniques. Finally, the presence of MAP was identified in 42.9% (3/7) of semen samples with nPCR; however, it was not detected through culture. In conclusion, the presence of MAP was identified in lymphatic, digestive tissue, and semen; the presence of MAP was reported for the first time in epididymis, Cowper gland, prostate and testicles of infected Pelibuey rams.

Abstract The production of squash (Cucurbita pepo L.) for vegetables or zucchini under cover presents difficulties due to the lack of pollination, for this reason, parthenocarpy is an alternative to generate greenhouse zucchini varieties, so efforts are currently being made in the search for genetic parthenocarpy that does not it requires the stimulation of pollination, nor by physiological factors in order that the fruit develops in a normal way until physiological maturity. A total of 46 parthenocarpic genotypes of round zucchini shrub type squash obtained at the Autonomous University Chapingo were studied. The objective was to characterize and identify the genetic diversity among the genotypes from the variability obtained by means of molecular genetic markers such as the microsatellites of the ISSR type (inter repeated simple sequences) and RAMP (microsatellite polymorphism amplified at random). It was used (GATA)4 as an ISSR initiator and the combination of (GATA)4 with four random sequence primers of the Operon series A (OPA05, OPA10, OPA13 and OPA19) for RAMP. With the two techniques, a total of 43 polymorphic loci were obtained, six loci amplified with ISSR and 37 loci with RAMP. The percentage of polymorphic loci for ISSR was 100%, while for RAMP it was 91.8% polymorphism, a high degree of genetic relationship was found between the partenocarpal genotypes. Five groups were formed for ISSR and four for RAMP, with gene distances between 0.72 to 1.

Resumen La producción de calabaza (Cucurbita pepo L.) para verdura o calabacita bajo cubierta presenta dificultades por la falta de polinización, por tal razón, la partenocarpia es una alternativa para generar variedades de calabacita de invernadero, por lo que actualmente se realizan esfuerzos en la búsqueda de partenocarpia genética que no requiera del estímulo de la polinización, ni por factores fisiológicos a fin de que el fruto se desarrolle en forma normal hasta la madurez fisiológica. Se estudiaron molecularmente 46 genotipos partenocarpicos de calabaza arbustiva tipo round zucchini obtenidos en la Universidad Autónoma Chapingo. El objetivo fue caracterizar e identificar la diversidad genética entre los genotipos a partir de la variabilidad obtenida mediante marcadores genéticos moleculares como los microsatélites del tipo ISSR (inter secuencias simples repetidas) y RAMP (polimorfismo de microsatélites amplificados al azar). Se utilizó (GATA)4 como iniciador de ISSR y la combinación de (GATA)4 con cuatro iniciadores de secuencia aleatoria de la serie A de Operon (OPA05, OPA10, OPA13 Y OPA19) para RAMP. Con las dos técnicas se obtuvo un total de 43 loci polimórficos, seis loci amplificaron con ISSR y 37 loci con RAMP. El porcentaje de loci polimórficos para ISSR fue de 100%, mientras que para RAMP fue de 91.8% de polimorfismo, se encontró alto grado de relación genética entre los genotipos partenocarpicos. Se formaron cinco grupos para ISSR y cuatro para RAMP, con distancias génicas entre 0.72 a 1.

Abstract. In a high-tech greenhouse located at Atlacomulco, Mexico State, incidences from 60 to 90% of plants showing symptoms associated to begomoviruses have been observed. The objectives of this study were to identify the begomoviruses present, to know if they are transmitted by seed, to determine the species of whitefly and if they are carriers of the virus. Leaves of adult symptomatic plants were analyzed with universal primers for begomoviruses. Seeds of fruits from positive plants were seeded to obtain seedlings; seeds and seedlings were analyzed by PCR for begomoviruses. Adults whiteflies were analyzed with specific primers to the mitochondrial gene of the cytochrome C oxidase subunit I and universal primers for begomoviruses. Sequences of PCR products from symptomatic plants had 99% of identity with Tomato severe leaf curl virus (ToSLCV). No begomoviruses were detected in seed and seedlings. Sequences from the whiteflies present in the greenhouse had an identity of 99% with Bemisia tabaci and they were carriers of ToSLCV. This is the first report of ToSLCV infecting tomato grown in greenhouse in Mexico.

Resumen. En un invernadero de alta tecnología ubicado en Atlacomulco, Estado de México, se han observado incidencias del 60 al 90% de plantas con síntomas típicos de begomovirus. Los objetivos del presente estudio fueron identificar begomovirus presentes, conocer si se transmiten por semilla, determinar la especie de mosca blanca y si ésta es portadora de los virus. Hojas de plantas adultas con síntomas se analizaron con oligonucleótidos universales para begomovirus. Semillas de frutos de plantas positivas a begomovirus se sembraron en almácigos para obtener plántulas, éstas y parte de las semillas se analizaron por PCR para detectar begomovirus. Adultos de mosca blanca se analizaron con oligonucleótidos específicos del gen mitocondrial de la subunidad I del citocromo C oxidasa y con oligonucleótidos universales para begomovirus. Las secuencias de los productos obtenidos por PCR de plantas con síntomas tuvieron una similitud de 99% con el Tomato severe leaf curl virus (ToSLCV). No se detectaron begomovirus en semilla y plántula. Las secuencias obtenidas por PCR de mosca blanca en el invernadero tuvieron 99% de similitud con Bemisia tabaci y los individuos analizados fueron portadores del ToSLCV. Este es el primer reporte del ToSLCV asociado a tomate en invernadero en México.

Abstract. In Montecillo Texcoco, State of Mexico we observed daisy plants (Dimorphotheca sinuata) showing symptoms of phyllody, virescence and proliferation of axillary branches, all those induced by phytoplasmas. Therefore, the objective of this investigation was to detect the phytoplasma associated with these symptoms by PCR with primers P1/P7 and R16F2n / R2 (nested PCR). The 1200 bp PCR product was obtained from the PCR and it was digested with the restriction enzymes MseI (Tru91), AluI, Kpn1 and HaIII and restriction maps showed that the phytoplasma present in Dimorphoteca sinuata is Candidatus phytoplasma asteris (16Sr1-B). The sequences were deposited on the NCBI data base and had a similarity of 99% with Candidatus phytoplasma asteris group 16Sr1-B from Iran (MH638316.1). A phylogenetic analysis was performed with the Neighbour-Joining method in which the phytoplasm from daisy was grouped with Candidatus phytoplasma asteris. According to the symptoms observed in the field, analysis of restriction patterns, sequencing and phylogeny, the phyllody of daisy is associated with Candidatus phytoplasma asteris, phylogenetically related to the 16Sr1-B group.

Resumen. En Montecillo Texcoco, Estado de México se observaron plantas de margarita (Dimorphotheca sinuata) exhibiendo síntomas de filodia, virescencia y proliferación de ramas axilares putativos a los inducidos por fitoplasmas. Por lo que el objetivo de esta investigación fue detectar el fitoplasma asociado a estos síntomas mediante PCR con los iniciadores P1/P7 y R16F2n/R2. Se detectó la presencia de fitoplasmas en tejido foliar sintomático. El producto de PCR de 1200 pb obtenido de la PCR se secuenció y sometió a digestión con las enzimas de restricción MseI (Tru91), AluI, Kpn1 y HaIII, los patrones de restricción evidenciaron que el fitoplasma presente en Dimorphotheca sinuata es Candidatus phytoplasma asteris (16SrI). Las secuencias obtenidas fueron depositadas en la base de datos del NCBI y tuvieron una similitud del 99% con Candidatus phytoplasm asteris grupo 16Sr1-B de Irán (MH638316.1). Se realizó un análisis filogenético con el método de Neighbour-Joining, en el cual el aislamiento detectado en margarita, se agrupó con Candidatus phytoplasma asteris. De acuerdo con los síntomas observados en campo, análisis de patrones de restricción, secuenciación y filogenia, indican que la filodia en margarita está asociada a Candidatus phytoplasma asteris, relacionado filogenéticamente al grupo 16Sr1-B.

Abstract The consumption of zapote mante (Pouteria campechiana) in Mexico has increased, being the municipality of El Mante, Tamaulipas, one of the main producers. In a non-commercial garden, sunken spots of brown color coalesced in fruits. The objective of this study was to identify the causal agent of the spot of fruit de zapote mante. With the fungi isolated, the Koch postulates were amplified, and the ITS region of the rDNA was amplified by PCR using the universal primers ITS1-ITS4. Morphological and molecular analysis indicated that the causal agent of the fruit blotch is the fungus Cladosporium cladosporioides. To our knowledge, this is the first report of this pathogen affecting this zapote mante in Mexico.

Resumen El consumo del zapote mante (Pouteria campechiana) en México se ha incrementado, siendo el municipio de El Mante, Tamaulipas, uno de los principales productores. En un huerto no comercial se observaron en frutos manchas hundidas de color marrón que coalescen. El objetivo de este estudio fue identificar el agente causal de la mancha del fruto de zapote mante. Con los hongos aislados se realizaron los postulados de Koch, se amplificó y secuencio la región ITS del ADNr mediante PCR utilizando los iniciadores universales ITS1-ITS4. El análisis morfológico y molecular indicó que Cladosporium cladosporioides es el agente causal de la mancha del fruto. Hasta donde se sabe, este es el primer reporte de este patógeno afectando a zapote mante en México.

Abstract The huitlacoche (Ustilago maydis (DC) Corda) is a good source of nutrients in the diet. During post-harvest handling, the product shows a rapid senescence, attributed to the high rates of dehydration and respiration of the product, in addition to its composition. In order to prolong shelf life, the effect of high relative humidity and low storage temperature on the physical properties of huitlacoche (whole and in galls) and the presence of phytopathogenic fungi were evaluated. The results showed that at 3 ºC the dehydration rate decreased, maintaining the visual quality for longer, both the whole huitlacoche and in galls. Storage with high relative humidity at room temperature favored the proliferation of phytopathogenic host species such as Fusarium oxysporum, Rhizopus stolonifer and Penicillium expansum. The highest incidence and severity was shown by F. oxysporum inoculated with a wound in the gall, while R. stolonifer, although with a lower incidence than the other two species, was able to survive up to 10 ºC. The results demonstrate that the management of huitlacoche at 3 °C and high relative humidity in ambient air is feasible, as it counteracts the rate of dehydration and eliminates the risk of proliferation of host phytopathogenic fungi.

Resumen El huitlacoche (Ustilago maydis (DC) Corda) es una buena fuente de aporte de nutrientes en la alimentación. Durante el manejo poscosecha, el producto muestra una rápida senescencia, atribuida a las altas tasas de deshidratación y respiración del producto, además de la composición del mismo. Con la finalidad de prolongar la vida de anaquel, se evaluó el efecto de alta humedad relativa y baja temperatura de almacenamiento sobre las propiedades físicas del huitlacoche (entero y agalla) y la presencia de los hongos fitopatógenos. Los resultados mostraron que a 3 ºC la tasa de deshidratación disminuye, manteniendo la calidad visual por más tiempo, tanto del huitlacoche entero como en agalla. El almacenamiento con alta humedad relativa a temperatura ambiente favoreció la proliferación de especies fitopatógenas hospederas como Fusarium oxysporum, Rhizopus stolonifer y Penicillium expansum. La mayor incidencia y severidad la presentó F. oxysporum inoculada con herida en agalla, mientras que R. stolonifer, aunque con menor incidencia que las otras dos especies, fue capaz de sobrevivir hasta 10 ºC. Los resultados demuestran que es viable el manejo del huitlacoche a 3 °C y alta humedad relativa en el aire del ambiente, ya que contrarresta la velocidad de deshidratación y elimina el riesgo de proliferación de hongos fitopatógenos hospederos de la superficie.

Abstract This paper provides an overview of the biological, ecological, epidemiological and management aspects of Candidatus Liberibacter, a bacterium recently introduced into Mexico that is associated with diseases such as citrus Huanglongbing, Zebra chip and chili pepper variegation. Plants affected with Ca. Liberibacter have multiple swellings located in the necrotic phloem and dispersed throughout the vascular system, massive accumulation of starch, aberrations in cambial activity and excessive formation of phloem. It is suggested that phloem necrosis causes blockage of the nutrient translocation stream, which leads to anatomical changes such as mottling and vein yellowing. Ca. Liberibacter control focuses on reducing inoculum sources, such as the use of healthy plants, removal of symptomatic tres.

Resumen En el presente manuscrito se proporciona una visión general de los aspectos biológicos, ecológicos, epidemiológicos y de manejo de Candidatus Liberibacter, bacteria de reciente introducción en México, la cual está asociada con enfermedades como: Huanglongbing de los cítricos, Zebra chip y variegado de chile. En plantas afectadas con Ca. Liberibacter se presentan múltiples abultamientos localizados en el floema necrótico y dispersos a través del sistema vascular, acumulación masiva de almidón, aberraciones en la actividad cambial y formación excesiva de floema. Se sugiere que la necrosis del floema causa el bloqueo en la corriente y translocación de nutrimentos, que conduce a los cambios anatómicos como el moteado y amarillamiento de venas. El control de Ca. Liberibacter se enfoca en disminuir las fuentes de inóculo, como el uso de plantas sanas, remoción de árboles sintomáticos y el control químico del insecto vector para reducir la transmisión de la bacteria.

Abstract: In El Bajo Papaloapan, the main producing area of pineapple of Mexico, leaves with typical symptoms of viral infection consisting in chlorosis, flaccidity, reduced growth and reddening were collected. By RT-PCR with specific primers for the hsp70 gene and subsequent sequencing were detected Pineapple mealybug wilt virus associated-virus 1 (PMWaV-1) and Pineapple mealybug wilt virus associated-3 (PMWaV-3). From the sequences obtained a tree was done with sequences from different regions of the world available in GenBank in order to know their similarity. The sequence obtained from the Mexican isolate PMWaV-1 was genetically related to the sequences of isolates from Cuba, Taiwan, Thailand and Hawaii and more distant from the Australian isolate. The sequence obtained for the Mexican isolate PMWaV-3 was more related to isolates from Hawaii, Cuba, Australia and Taiwan and more distant from the Thailand isolate. This is the first report of the presence of these two viruses in Mexico.

Resumen: En El Bajo Papaloapan, principal zona productora de piña, se colectaron hojas con síntomas de clorosis, flacidez, reducción del crecimiento y enrojecimiento foliar típicos de infecciones virales en este cultivo. Mediante RT-PCR con indicadores específicos para el gen hsp70 y posterior secuenciación, fueron detectados los virus Pineapple mealybug wilt associated-virus 1 (PMWaV-1) y Pineapple mealybug wilt associated-virus 3 (PMWaV-3). A partir de las secuencias obtenidas se construyó un árbol con secuencias procedentes de diferentes regiones del mundo disponibles en el GenBank para determinar su similitud. La secuencia obtenida del aislamiento mexicano del PMWaV-1 resultó ser más próxima genéticamente a secuencias de aislamientos procedentes de Cuba, Taiwán, Tailandia y Hawai y más distante del aislamiento australiano. La secuencia obtenida para el aislamiento mexicano del PMWaV-3 se encontró más relacionada con los aislamientos de Hawai, Cuba, Australia y Taiwán y más distante del aislamiento tailandés. Este es el primer reporte de la presencia de estos dos virus en México.

En 2010, dans la région de Nopaltepec, État de Mexico, ont été détectés des plants de figue de barbarie présentant des symptômes de déformation et grossissement du cladode, mosaïque, jaunissement et déformation des fruits sur tout ou partie de la plante. Le syndrome a été associé à une infection occasionnée par phytoplasmes. Le but de cette recherche a été d'identifier par réaction en chaîne par polymérase (PCR) la présence de phytoplasmes dans différentes strates du cladode et dans la racine des plants de figue de barbarie. L'ADN extrait d'échantillons de plantes présentant des symptômes et de plantes asymptomatiques, a d'abord été amplifié avec des amorces universelles P1/P7 et amorces P1/Tint, puis par PCR nichée en utilisant des amorces R16F2/R16R2. Un fragment de 1200 pb a été obtenu dans tous les échantillons avec les symptômes ci-dessus. L'analyse des profils de restriction générés par les endonucléases HaeIII, KpnI et MseI (TrU91) a révélé que le phytoplasme appartient au groupe 16SRXIII-virescence de la pervenche mexicaine (mexicaine periwinkle virescence).

During 2010 in Nopaltepec, Mexico, cactus plants were detected with symptoms of distortion and thickening of the cladode, mosaic, yellowing, proliferation and deformation of fruits in the whole plant or part of it. Since the syndrome has been attributed to phytoplasma infection, the objective of this study was to detect, the presence of phytoplasmas in different strata of the cladode and the roots of cactus plants by Polimerase Chain Reaction (PCR). The DNA extracted from cladodes with symptoms and asymptomatic, was amplified firstly with universal primers P1/P7, and P1/Tint, fragment of phytoplasma 16S rRNA gene, followed by nested PCR using the primers R16F2/R16R2, a fragment of 1 200 bp was obtained. Analysis of restriction patterns generated with endonucleases revealed that the phytoplasma belongs to the Mexican periwinkle virescence-16SrXIII.

Durante 2010 se detectaron en la zona tunera de Nopaltepec, Estado de México, plantas de nopal con síntomas de deformación y engrosamiento del cladodio, mosaico, amarillamiento, proliferación y deformación de frutos en toda la planta o en parte de ella. Dado que el síndrome se ha relacionado con la infección por fitoplasmas, el objetivo de esta investigación fue detectar mediante la Reacción en Cadena de la Polimerasa (PCR), la presencia de fitoplasmas en diferentes estratos del cladodio y raíz de plantas de nopal. El ADN extraído de las muestras de plantas con síntomas y asintomáticas, se amplificó primero con los iniciadores universales P1/P7 y P1/Tint, y posteriormente mediante PCR anidada utilizando los iniciadores R16F2/R16R2, se obtuvo un fragmento de 1 200 pb en todas las muestras con los síntomas antes mencionados. El análisis de los patrones de restricción generados con las endonucleasas HaeIII, KpnI y MseI (TrU91), reveló que el fitoplasma pertenece al grupo 16SrXIII-Mexican periwinkle virescence.

Le piment (Capsicum annum) CM334 est résistant à l'oomycète Phytophthora capsici (Pc), cette resistance se manifestant par une réaction d'hypersensibilité. Cependant, ce génotype présente une sensibilité à Fusarium oxysporum (Fo). Dans cette étude, nous avons comparé les changements cellulaires dans la réaction d'hypersensibilité induite par Pc (l'interaction incompatible), et la nécrose induite par Fo (l'interaction compatible). La nécrose et la colonisation occasionnées par l'oomycète dans l'interaction incompatible ont été limitées au site d'inoculation dans la tige. Dans l'interaction compatible, l'agent pathogéne a colonisé toute la tige et la nécrose a été limitée au site d'inoculation. La dégradation des parois cellulaires était évidente dans l'interaction compatible, alors que dans l'interaction incompatible a été observé l'effondrement des cellules. Dans l'interaction incompatible 31% des noyaux des cellules corticales étaient rouges, suggérant une réaction d'hypersensibilité. Dans l'interaction compatible 71% étaient globuleux et gris, suggérant la mort par nécrose.

The chilli (Capsicum annuum) CM334 is resistant to oomycete Phytophthora capsici (Pc), such resistance is manifested by a hypersensitive reaction (HR); however, this genotype is susceptible to Fusarium oxysporum (Fo). In this study we compared the cellular changes in the HR-induced by Pc, incompatible interaction (II), and Fo-induced necrosis, compatible interaction (CI). The necrosis and colonization by the oomycete in II was confined to the site of inoculation in the stem, and in CI, the pathogen colonized the whole stem and the necrosis was limited to the site of inoculation. Cell wall degradation was evident in the CI, whereas in the II, only the collapse of the cells was observed. In the II, 31% of the nuclei of cortical cells were red suggesting a HR and in the CI 71% of the nuclei were globose and light gray suggesting a death by necrosis.

El chile (Capsicum annuum) CM334 es resistente al oomiceto Phytophthora capsici (Pc), tal resistencia se manifiesta por una reacción de hipersensibilidad (RH); sin embargo, este genotipo es susceptible a Fusarium oxysporum (Fo). En el presente trabajo se compararon los cambios celulares en la RH inducida por Pc, interacción incompatible (II), y la necrosis inducida por Fo, interacción compatible (IC). La necrosis y la colonización por el oomiceto en la II se confinó al sitio de la inoculación en el tallo; y en la IC, el patógeno colonizó todo el tallo y la necrosis se limitó a la zona de inoculación. La degradación de las paredes celulares en la IC fue evidente, en cambio en la II, solamente se observó el colapso de las células. En la II el 31% de los núcleos de las células corticales fueron rojos sugiriendo una RH y en la IC el 71% fueron globosos y de color gris claro sugiriendo muerte por necrosis.

A subtractive library of sequences associated to proteins synthesis in hyphal germination of Sclerotium cepivorum was generated. In order to generate the subtractive library, optimal conditions to have eruptive and hyphal germination of S. cepivorum sclerotia were determined. At 22° C, 96.7% of sclerotia germinated hyphally without allyl disulfide (DAS) and 91.67% of sclerotia germinated eruptively in presence of DAS 2%. Twenty three fragments of genes associated to hyphal germination of S. cepivorum were obtained in the subtractive library. Those fragments had significant similarity to 15 different proteins that participate in respiration, transcription, translation, signal transduction and amino acid and protein synthesis process. The results suggest that hyphal germination is faster than eruptive germination because have an increment in the metabolic processes in sclerotia of S. cepivorum germinated hyphally.

Se generó una biblioteca sustractiva con las secuencias asociadas a la síntesis de proteínas durante la germinación no errumpente de Sclerotium cepivorum. Para generar la biblioteca sustractiva se determinaron las condiciones óptimas para obtener germinación errumpente y no errumpente de esclerocios de S. cepivorum. A 22° C, el 96.7% de los esclerocios germinaron de forma no errumpente en ausencia de disulfuro de alilo (DAS), y el 91.67% de los esclerocios germinó de forma errumpente en presencia de DAS al 2%. En la biblioteca sustractiva se obtuvieron 23 fragmentos de genes asociados a la germinación no errumpente de S. cepivorum. Estos fragmentos tuvieron similitudes significativas con 15 proteínas diferentes, las cuales participan en los procesos de respiración, transcripción, traducción, transducción de señales y síntesis de aminoácidos y proteínas. Los resultados sugieren que la germinación no errumpente es más rápida que la germinación errumpente debido a un incremento en los procesos metabólicos de los esclerocios de S. cepivorum germinados de forma no errumpente.

Potato purple top affects yield and tubers quality by browning and the abnormal sprout associated with it. This disease is considered of phytoplasmic origin, although Koch postulates have not been properly proven. This investigation tries to determine etiology for browning and abnormal sprouting in potato; taking this into account isolations coming from 19 genotypes of field potato with four varieties were transmitted by means of graft: Sangema that had 52.63% transmission, Michoacán that had 42.1%, Norteña with 31.57% and NAU-6 that reached 65.2%. The greenhouse potatoes cuttings with symptoms of purple tip transmitted by seed-tuber, induced symptoms of "popotillo" in red tomato; however, when grafting again in potato symptoms were not observed in the foliage, neither browning in the tubers and phytoplasma was not detected by means of reaction technique in polymerase chain. The transmission percentage for potato purple tip in greenhouse was of 37%. The browning and the abnormal sprouting in tubers were induced by an infectious pathogen, transmitted until for six successive grafts in greenhouse. The percentage of phytoplasma detection with polymerase in foliage and tuber of used varieties NAU-6, Norteña, Michoacán and Alpha were of 37.5, 10, 35.7 and 28.6 respectively. In chili manzano and mixquic, this was of 70.83% and 66.6%, in tomato there were not recorded phytoplasmas. In chili plants, it can act as intermediary host in transmission of pathogen involved with this disease.

La punta morada de la papa afecta el rendimiento y la calidad de los tubérculos por el pardeamiento y la brotación anormal asociados con ésta. Esta enfermedad es considerada de origen fitoplásmico, aunque los postulados de Koch no han sido debidamente probados. Esta investigación intenta determinar la etiología del pardeamiento y la brotación anormal en papa; para ello, se transmitieron mediante injerto, aislamientos procedentes de 19 genotipos de papa de campo con cuatro variedades: Sangema que tuvo 52.63% de transmisión, Michoacán que tuvo 42.1%, Norteña con 31.57% y NAU-6 que alcanzó 65.2%. Los esquejes de papas de invernadero con síntomas de punta morada transmitidos por semilla-tubérculo, indujeron síntomas de "popotillo" en jitomate; sin embargo, al volver a injertar en papa no se observaron síntomas en el follaje, tampoco pardeamiento en los tubérculos y no se detectó fitoplasma mediante la técnica de reacción en cadena de la polimerasa. El porcentaje de transmisión para la punta morada de la papa en invernadero fue de 37%. El pardeamiento y la brotación anormal en los tubérculos fueron inducidos por un patógeno infeccioso, trasmitido hasta por seis injertos sucesivos en invernadero. El porcentaje de detección de fitoplasma con polimerasa en follaje y tubérculo de las variedades utilizadas NAU-6, Norteña, Michoacán y Alpha fue de 37.5, 10, 35.7 y 28.6 respectivamente. En chile manzano y mixquic, éste fue de 70.83% y 66.6%, en tomate no se registraron fitoplasmas. Las plantas de chile, puede actuar como hospedante intermediario en la transmisión del patógeno involucrado con esta enfermedad.
